有人用过Tet-pLKO-puro （shRNA vector）做 inducible protein overexpression 吗？# Biology - 生物学

没看到add card option. 还是一张卡一个帐户?

我把一个蛋白的基因插在本来是shRNA的位点（AgeI site），做了transient
overexpression，可以做puromycin筛选，检测到蛋白表达，但是做了virus之后，加
puromycin，细胞全死了，说明puromycin没有表达。这是为什么呢？BTW，在我的基因
前加或者不加promoter，会有什么影响吗？

应该有一个Link an Account的选项，点这个就行了

看了下cell cycle 2009 原文，这个系统用的inducible H1 promoter,应该属于PolIII
，不能转录蛋白基因。你瞬转检测到表达，有可能是从lenti vector的external promo
ter转录的（我猜）。包装病毒后没有这个promoter了，所以没表达。
但是你说puro都不表达了，那可能是你病毒都没包装成功。
建议换载体。

谢谢你的解释！我可以问一下，为什么包装病毒后就没有H1 promoter了吗？另外，病
毒包装不成功是受了插入的这个基因什么影响吗？
是在考虑换载体。

pLKO.1不太好改造，插入片段位置不恰当经常会影响病毒包装。

病毒系列的貌似Rudolf Jaenish用来做可诱导iPSC那个FUW-TetO-Oct4/...系列应该可
以，很多实验室都重复过，不过我没有亲自用过，另外这个系统rtTA没有做进FUW-TetO
里，你需要同时感染表达rtTA的病毒。
