currently no appointments available!"。到这一步为什么没法“继续”了?

发现一个转录因子,想做些工作。该因子的插入突变导致物种很明显对一种矿质营养元
素的缺乏很敏感(与wild-type比较),GFP 定位表明在核内,理论上该转录因子不大
可能直接调节质膜上的离子通道,一般是通过结合到某些下游基因的启动子区域而调控
下游基因的转录。打算先用芯片先找出哪些基因的表达受影响,不出意外的话,会有一
些靶基因(间接+直接的)出来,怎样才能找到关键的一个或某个基因,而该受影响的
基因就是导致离子吸收或转运出现异常的。 换句话说,怎样pinpoin该转录因子关键下
游靶基因呢?

By using expression assay, a lot of candidate genes will be identified
whose expression levels are regulated by this transcription factor. These
candidate genes should be further verified by real time PCR and western
blot. You can also use ChIP-seq assay to demonstrate the transcription
factor directly binds to the promoter region of some of the target genes.
However, the most important experiment to test whether this gene is the
real target of that transcription factor is a rescue experiment. I
whose expression levels are regulated by this transcription factor. These

I vaguely remember that I saw some cool drug target identification methods
published early this year, but I don't remember the details. It seems to
be drug affinity chromatography based for target identification. You
probably can google it. If you couldn't find it, post back here, others
might know. good luck!

be drug affinity chromatography based for target identification. You
