最近在做蛋白纯化，用的zymogen his6 column，elution完后，用SDS-LB煮了5分分钟
，发现western有若干条带。
实验室的人说是imidazole会降解蛋白。在网上查也有不少人说imidazole会降解蛋白。
请问各位是否遇到这样的问题。

没见过imidazole降解蛋白。要排除在细胞内就降解了（或者修饰变大了），或者抗体
有问题。

谢谢。网上说的倒不是imidazole会降解蛋白，而是imidazole在高温和SDS-LB煮的时候
会降解蛋白。问了周围的人，他们一般都dialysis。不知道synbio79有没有啥好推荐的
dialysis kit。

一般都会有些污染吧，没可能是非常纯的。只要目标蛋白足够富集就好了

in your case,
1. target protein is degraded before chromatography
or
2. 6his affinity chromatography can not purify it well
dialysis does not help in both
to determine if #1==TRUE, western blot using target specific antibody
to improve #2, add one more chromatography (ion exchange, size exclusion,
etc)

Thanks a lot.The problem is we don't have antibody for the protein.I am
using the second tag to tandem purify it and will see.
