A question about making knock-in mice through traditional homologous
recombination---After identifying the positive ES clones by southern blot
analysis, do people usually sequence the whole new allele from 5' arm to
the end of 3' arm? Assume that the targeting vector has been fully sequenced
before electroporation.

Sometimes the ends can be screwed up during recombination, so I would PCR
the arms out using external/internal primers and sequence then, just to be
safe.
