I want to try both NIW and EB1b, but do not want bother the reference to
write two letter. So, just want to know if one remmendation letter can use
for both NIW and EB1b ?

not quite... 讨论research的部分可以相似，但是NIW强调对美国的
national interest, EB1b强调outstanding in the field,
international recognition.
