ABSTRACT:
To investigate optimal parameters for preparation of BSA-loaded
nanocomposite microparticles (NCMPs) using biodegradable polyester poly (
Glycerol Adipate-co--Pentadecalactone), (PGA-co-PDL), by the double
emulsion solvent evaporation method followed by spray drying and
characterize their properties. The experiments were performed according to
Taguchi design. The effects on the particle size, BSA encapsulation
efficiency and loading were investigated. Optimized nanoparticle
preparations were further processed by spray drying to produce BSA-loaded
NCMPs. NCMPs were examined for their morphology and yield and optimized
preparations were further examined for their aerosolisation, in vitro
release behavior and toxicity. The size was mainly affected by the polymer
mass and small particle size ≤ 500nm achieved. While most important factors
for obtaining high BSA loading was BSA concentration. The spray drying
process was optimized to produce NCMPs with a porous corrugated surface and
48% yield. The identified optimum conditions of spray drying (aspirator
capacity 100%, Inlet temperature 100°C, air flow 400l/h, feed rate 10% and
total solid concentration 12.5mg/ml) produced NCMPs with an aerodynamic
particle diameter 1.46±141 µm (p>0.05, ANOVA/Tukey's) considered
suitable for inhalation (FPD 45±4.7µg and %FPF 78.57±0.1) and
cumulative BSA release % 62±4.7 after 48h. These results enable optimal
process parameters for preparation of BSA-loaded NCMPs suitable for
inhalation.
