To detect a 250KD protein, what concentration of SDS-PAGE gel should I run?
Do I need a stacking gel? If yes, what concentration?

actually, 10%-12% is fine. Stacking gel is always better.
When I run 400kd protein, I use precast gel. If you have precast gel, it is
good for any protein

buy precast , 4-12% is good for this silze
if u make it by yourself, I will say 6% or 8% is good
the protien band will show up on any percentage gel( by adjusting running
time), the real issue is how well it can separate contaminant next to it
from your protien.

6%-7% running gel. You still need a stacking gel (2%-4%)
