I am thinking that ,theoretically, if the male gender ES cell are injected
and subsequently make it to germ line contribution, the chimera pups tend to
be in favor of Male gender type.
But, I don't know if it is true, anyone here who is more experienced on this
topic can tell me whether I am right or not ?

If the ES cells contributed to germ line, they should be able to convert
the gender type of the chimera pups. In another words, good Chimeras should
be male.

I am having the same problem: some people here said that high agouti combine
with distorted sex ratio is usually a guarantee of successful germ line
transmission
but I am waiting for it to really happen -_

Which cell line are you using ? I saw some literature mentioning that G4 or
Sv129 derived line is much more robust and with predictable behavior than C57 derived lines.
