Need to separate some virions with Sucrose ultracentrifugation (PI said we
don't need to do CsCl :( - that would have been easier to search for
literature if so). Anyway, I did some trial runs. My goal is to separate the
samples to several layers (if there is different shape of viral particles).
Then collect the layers and run on western blot to see and compare.
take I : 17300 rpm at 4C, x 90 min --> no separation
take II: 30000 rpm at 4C, x 90 min --> no separation
take III: 30000 rpm at 4C, x 3 h ---> still no separation
the sucrose pads i used are: 20% (upper), 35% (middle); 50% (bottom); let
sit at 4C overnight before loading samples and ran the program.
total volume 2 ml (each layer 0.5 ml -- including sample layer).

rate of your virion, you can have an educated guess about the conditions you

我的理解，如果只在多伦多或者温哥华转一次，不需要
只要不是这样都需要
另外，如果经济宽裕的话还是别那么折腾母亲了，买个直飞或者转一次的吧

这个也太折腾父母了，花点钱换个直达的吧. 两次转可能要签证，因为从
international到
domestic再到international. 你父母如果不懂英文，从international到domestic再到
internation很折腾, 而且可能误点。

不需要,但是订座时需要请人加注,过境不需要签证
