What's the weed? Hopefully it isn't Bermuda :-)
Right, round up once probably isn't enough. When people do lawn renovation,
they round up once, then water for a few days to a week, and round up once
again. Sometimes people do it a third time. Still, this won't guarantee
there won't be weeds in the new lawn, just much fewer.
If you don't like roundup, you can try organic solutions like vinegar (15-20
%, dangerous though), baking soda (spray powder form after spraying some
soapy water), or boiling water. Note they are non-selective, which means
they kill good grass too. Still, they won't guarantee no new weeds will
grow back.
So it is basically a continuous process, provided there is organic matter,
dirt, moisture and light.
Also don't add wood mulch too close to your foundation for termite concerns.
If you have to, at least 6"-12" a gap should be the norm.
