to study an unknow gene, u need northern first ahead of RT-PCR.
until you have a very specific question about a gene, which
has been studied for a long time.
there are several reasons:
(1) the distribution of a gene
(2) the abundance of a gene
(3) possible isoforms of a gene
such as a simple question is here: the the gene is 3 kb or longer,
u can easily meet a lot of PCR problems to amplify it.

I guess, you may misunderstand what your director's idea.
He may wanna know before identification if the transcription level has
significant difference between the WT and mutants. For people always care
much a functional mutation rather than a sequence mutation itself. And most
researchers like to start from a functional change. Your idea could be
classified into reverse genetical method. It is ok, but not often be used in
a single gene case.
