今天我从我15岁的女儿电脑上搜出来一部A片，藏在“未来的职业”文件夹里，未来的
职业！What a

晚上我女朋友说我太娘了，我很火大，就跟她吵起来了，本来是想显得男人一点，结果
最后控制不住
哭起来了。What a fucking day!
昨天晚上我裸着就穿一内裤躺床上，边喝酸奶边上网，不小心酸奶滴到身上了，我正在
擦，我爸突然
进来了，居然以为我在SY什么话也没说扭头又关门出去了。What a fucking day!
今天对着镜子端详了一会儿，突然发觉自己长得有点像李宇春！What a fucking day!
周五喝多了带一女的回家，没有套，我有点担心搞出事，那女的说没事，及时出来就行
了。结果等到
高潮我控制不住了正要拔出来的时候，那女的突然两腿一盘裹住我的腰，死命夹着我大
叫：“当我孩子
的爹吧！！！” 我当场就在里面完事了。What a fucking day!
新买了件情趣内衣，昨天晚上吃完饭了悄悄换上，爬到饭桌上挑逗老公。。。。。。。
。。。。。。
桌子塌了，负伤，内衣撕破。What a fucking day!
今天和我小外甥坐沙发上一边吃

一月份买了一个WNDR3700，用的WPA-TKIP加密，无论笔记本带的Intel 5100还是台式机的
Linksys WMP600N PCI网卡都只能连到54MBps，相反，一片老得不行的Belkin PCMCIA卡
倒是能
连到270-300Mbps，信号强度没问题，满格或者接近满格。
一直到不到问题所在，最近想到看看网卡的技术参数，发现两片只能连到54M的网卡只
支持WPA2加密，而
不正式支持WPA加密，把加密改成WPA2，速度立马上到270-300，如果大家碰巧也有这个
情况，可以试
试。

本人对siRNA的设计和应用几乎无知，问题比较菜，请大家不要见笑。
需要用siRNA来knockdown一个外源表达的fusion protein(GFP-GOI)，
因为不想影响endogenous GOI，所以想请教能不能设计siRNA只针对GFP，而不是GOI？
这样的siRNA会有效果吗？
另外大家都用什么program来设计siRNA，另外在哪家公司买siRNA啊？

I guess you can silence GFP-GOI transgene by only targeting GFP region
without affecting endogenous GOI expression.

can you please explain more why targeting GFP only does not
interfere with the translation of the fusion protein?

if this works, that would be exactly what I want.
in this way, I can determine if the phenotypes
are induced specificly by GFP-GOI transgene.

外源表达的干嘛需要knockdown啊？
你不转到细胞里不就行了么？
要用siRNA knockdown GFP-GOI而不影响GOI表达当然可以做到，设计target GFP序列的
siRNA就可以了。

他应该是想用siRNA去除GFP-GOI to verify their GFP signals (localization
pattherns, maybe?).

we want to determine if the phenotypes of the transgenic animals
are specifically caused by GFP-GOI transgene.
if by knocking down GFP-GOI transgene only, but not endougeneous GOI,
the phenotypes are gone or less severe, then we are more confident that
ectopic expression of GOI induces the phenotypes.

In theory it should work, but then you have to make sure it is not due to
off-target effect, and later rescue the phenotype by making RNAi resistant
GFP-GOI to demonstrate specificity. What's more, what if the effect of
endogenous and ectopic GOI are not additive or synergistic?
