是不是G418只能筛选HEK 293？HEK 293T自带G418抗性？

HEK 293T 本身不带G418 抗性。
但是因为 293T可以不停的复制环状的质粒，所以不管是否稳定转染，都会有抗性，
会导致你很难筛选出被稳定转染的细胞。

如果你的问题是：一个已经整合了lenti-vector的293T细胞，再转进packaging mix，能
不能自己produce virus?
这个肯定不行的。你仔细看下lenti vector的结构就知道了，vector是有external pro
moter能转录整个viral RNA，而整合以后的片段不行。这也是safety的设计，在第三代
lenti viral vector的LTR自己没有转录活性了，叫做self inactivated。

你是说带有sv40 ori的质粒能被T antigen复制吧
？

293T本身应该是G418 resistant，不然它那个大T抗原怎么进去的。

Oh...I thought it was accidentally made with antigen T by virus infection or
something. Guess I was wrong.
So it was intentionally made by stable transfection?
