一般都是normalize到house-keep genes,对吧。
你们见过normalize到ponceau S staining的吗?
读了篇paper,里面是那样弄的,
转完膜,Ponceau S染,然后记录下来,说为了保证equal loading,
然后再用抗体得到specific signal, normalize到corresponding Ponceau S signal。
。。
我从来没有见过这么做的,也觉得这么做是不对的。
大家的意见呢?

怪怪的,他这相当于用总蛋白normallize吧
虽然我觉得大多数情况normalize的结果和tublin 之类housekeeping gene的Normalize
结果也差不多,有时还比housekeeping gene的 normalize结果更准确些……

最近也在看这些技术文章,似乎如果染料结合没饱和的话,这么做也不一定比house
keeping genes差。前不久看过的一个全蛋白做normalization文章:

另外还有个半定量western的文章提到Ponceau S的局限
It has been found that images acquired with fluorescent staining are better
suited for quantification compared to Ponceau S. This is connected to the
fact that a color staining such as Ponceau S darkens the membrane locally.
Since the color of the membrane cannot become darker than that of the pure
staining color, there must be saturation before that color value is reached.
This leaves only a certain useful range of staining concentrations,
independent of whether the actual binding of Ponceau S to the protein is
saturated. With fluorescent staining, the membrane appears brighter with
increasing protein concentration, and there is no theoretical upper bound
for brightness.
