(不信神、不信教的请绕行，不必回帖，话不投机半句多)
找了几位看看，都是2016年经过考验、在川普面对KHAN门、抓猫门形势不利时仍
然相信川普赢的那种水平的算命先生。
1. 川普是上天派来干大事的，人类过去被锁住的科技会在川普任上缓慢解封。
2. 新技术包括航天科技、新能源(零点能量)、反重力技术、医疗科技等等。
3. 新科技革命。以Elon Musk为首的一批公司会开始逐步推广新技术。
4. 全球都有份，中、俄、印、欧等地区也会得到相应适用的科技。
5. 亚特兰提斯时代这些科技就给人类了，但当年人类玩火走向自我毁灭。作为
前车之鉴亚特兰提斯后人类工程重启这些技术被严控。51区、NASA等机构早就
从外星人得到这些技术但不得外泄，直到人类成熟到一定阶段才可以。
6. 和外星人的接触开始变明朗，但不是通过官方渠道，而是各种LEAK。
个人感觉影子政府(Dark Illuminati)对此甚反感，会对川普施加负面影响。
如果川普不为所动上述几条都可以实现，否则还要再等等。

我们做microarray发现了一些基因表达降低，随后用real time PCR 也得到确认。但我
们试图用western blot及ELISA的方法去证明蛋白表达也降低时却遇到麻烦，因为
western和ELISA显示蛋白表达正常。有点想不通。大家遇到过这样的情况吗？

of real time PCR. 但在这里好像不是这么回事。

很正常，我碰到过某蛋白mRNA调控上升明显，westernblot出来是急剧减少，还以为实
验哪出问题了。troubleshooting了很久，发现就是这么回事。。。

do you know the half life-time of your interested protein? Also there could
be other post-transcriptional mechanisms involved.

you described will happen. I heard that usually the WB results match with
real time PCR. Is that right?

在Harvard开始的方向是在单细胞+单分子水平上观察细菌基因的表达和调控，主要是分
析Transcription和Translation的随机性。有一个结果特别有意思，就是在大肠杆菌里
面，任何一个时刻mRNA的copy number和对应蛋白质的copy number完全没有任何相关性
。这个方向成就了好几个现在转做系统生物学的人。

try below steps:
IN VIVO sampling from cell lines/KO^+ + or KO^-- Mouse
separated to half and half
with half do mRNA microarray and real time PCR
then await 12 hr another half do WB. NB: this 12 hr should be cultured at
same cell line/Mouse

usually the protein does not match mRNA . It's normal.
