请教大家几个问题
1、 选puromycin浓度的时候，需要提前做个proliferation 的试验来确定dose，大家
treat cells几天，是选80%抑制的dose么？
2、 作colony formation assay的时候，因为做的时间比较长，等的过程中，溶液中重
要加药物筛选么？

1、puromycin作用时间有时候会比较慢，视细胞株而异，建议做1个礼拜左右观察。建
议使用100%抑制的最低浓度，否则很可能混入假阳性克隆。
2、我一般3-5天换一次液。
