没必要自己找事，没人会知道。

这是个方法，值得考虑。但是很多细胞系over-express GPCR还是很困难的。而且即使
不成功也不能完全排除Gs alpha的可能，因为也许还有其他必要signaling molecule被
over-express 这个GPCR disrupt了。
有没有更好的方法，最好能provide smoking gun这种。我自己有些想法，但是还是希
望集思广益一下。

哇靠，这个问题超过了我的专业范围，我只能大概就我知道的其他文章的例子
来建议一下吧。
要检测是否通过G protein, 无非就是这么几个思路：
1. G-protein specific inhibition: non-hydrolyzable GTP analog, GEF analog,
Knockdown of G-protein.
2. G protein dominant negative mutant
3. G protein dominant positive mutant
4. Or if you know something is in the middle of a pathway, you can also cut
off that middle player with methods similar as above.

dominant positive mutant需要做screening 的，除非你有这个资源，不然不要
自己做。一般就是直接查文献看看你关心的那个蛋白有没有人做过。
大体思路就是，其实GTP什么的就是用电荷来改变蛋白结构嘛，所以干脆把相关
binding pocket 的氨基酸给突变加上负电荷，有时候是可以产生dominant positive

另外，non-hydrolyzable GTP analog和GDP analog，你难道不能用脂类包埋的
方式来辅助过膜么？要确认是否成功过膜也比较简单，同时放点可溶的荧光分子
就好了。

试过，不work。文章里有先permeablize cell的，但觉得动作太大了，会引起很多
artifact，尤其我们要看的readout还很慢。

那就比较麻烦了。
先看看有没有人做过你们用的Ga的突变体吧。
另外，要证明是否artifact 还是可以的，关键是要做好control experiments,
比方说用普通的GTP 进行对照。我也知道有人用microinjection 的方法来做的，
但是这个方法一样是artifact非常严重，关键是要作好对照。

我们以前就是inject的，但是readout很快，而且是单细胞readout,作了很多control。
用permeablized的方法主要担心细胞活不了这么久。。。

在我听来的印象中，permeablize 的方法并不比injection harsh 多少。

也许。但是之前是inject后几分钟就可以看到变化或者做测试，这里要几十个小时。。。
