Don't forget, cellular ATP source is from the adenine pool. Some cells has
90% of adenine in ATP normally. Therefore, how can you increase your ATP
level by 10-30 folds? How about you just do the ATP step? Did you check
your standard curve?
Also, if you have ATP increases, why your cells are died?

90% of adenine in ATP normally. Therefore, how can you increase your ATP
level by 10-30 folds? How about you just do the ATP step? Did you check
your standard curve?
Also, if you have ATP increases, why your cells are died?

I doubt this results too, but the data is repeatable, so I want to try
another assay to confirm it.

You ATP kit is the luciferase kit. If you want stable signal try this kit (
less sensitive but stable):

Btw, the gold standard for ATP/ADP/AMP determination is the HPLC method.

个人觉得至少要做一个ATP／ADP ratio
最好能有ATP／ADP／AMP的ratio
