Here is my situation. My company will pay the closing cost. I got a letter
from the relocation company regarding relocation expense reimbursment. It
said that "the expense cut-off date is set at Nov 30, 2010. If you were
authorized for home purcahse closing costs and the expenses are not
finalized by Nov 30, any income based on this benefit will not be reported
in 2010. The deduction points in Tax Year 2010 for income that has not been
reported and the subsequent reporting of that income the following tax year
may have an adverse impact on you".
I can close either at the end of November or Dec 8. One friend said I should
close the later the better. For tax purpose, which date is better? If I
close on Dec 8 and report the expense after that, what adverse impact will

就差8天，在tax上有很大收获么。
我建议在11月底close，那样prepaid interest charge的少，从12月份收到的bill就已
经开始还本付息了。如果是12月8日close，那么银行会先charge 8日到月底的interest
，然后从1月的账单开始还本付息。对于closing当月，会不会按天算应还的本金，还没
有仔细研究过。

原核里面off-target 比例也类似么？ 弄一堆容易出现的off-target序列，放在生存
必须的基因里，但是target的序列条件致死（比如和一个筛选因子结合后致死）。然后
error-prone PCR crisper系统 电转不知
道可以不。。

昨天俺刚刚跟postdoc说了这么个project.
不过这东西拿grant也不容易，最好是让风投来做一个startup.

我觉得CRISPR的特异性应该还行，至少从上述的Paper里20个碱基，只要3-4个以上有变
异，off target效应就急剧下降。

做细胞的话多选几个克隆，设计2－3个CRISPR，得到一致的表型就行了
做动物的话，得到杂合体后back cross几代应该就没事了

要这么麻烦的话，为何不用其他方法，反正都能搞出来，就看哪个时间成本低

ankyrin说的对。其实siRNA和shRNA同样有这个问题，任何技术都不是万能的。
