签了一个foreclosure bank-owned house。在home inspection后发现一些不大不小的
问题。请问如果拿着contractor的quote和银行再negotiate价格或者要credit，银行接
受的可能性大不大？
房主2006年买入价是386K,房子银行287K foreclosure买入,给我们的价格是270K.另外
，seller agent和银行都非常想把这个房子赶快卖掉.

我要在细胞中表达一个Fe-S cluster SAM的蛋白， 这个蛋白活性对oxygen极为敏感，
至少有氧的情况下在E.coli中没有任何活性。
第一次把此蛋白在细胞质中表达，没有酶活。想问大家如何有何方法让此蛋白在细胞中
有酶活？把它在mitochondria中表达， 是否有帮助？

厌氧没法做？不然细胞里或多或少有氧气吧，加上你的是极为敏感。。。

这个估计只能在无氧条件下提，
还原态的Fe才是有活性的，O2是底物之一吧？你的铁已经是三价了，失去了被氧化的能
力，也就没有活性了。
如果没有无氧的条件，试试看提纯过程中加入适量DTT之类，同时把Buffer都Degas一下
。测活的时候再输入额外的O2。

如果是在yeast等耗氧厉害的host做
可以直接加到厌氧管 密封好
少量氧气很快被消耗掉 就成厌氧环境了
用这个至少pfl可以功能表达 我们做过 其他类型protein的就不知道了

sold 'as is'? bank normally will not do anything for the repairing and they
put it up front on the contract.
