我在切胚胎脑组织，很小。我是先4%PFA固定overnight，然后PBS wash 3 times, then
put in 30%sucrose overnight。Then put the tissue in OCT. Then quick freeze
by putting the cryomold in isopentane with dry ice.Cryosection at -20 degree
, cut 10um.切过后发现组织中总有裂痕，空洞，有时象是被撕开一样。请问怎么避免
