板上有养小鼠T细胞的大虾吗？
自己从小鼠脾脏分离的T细胞，用RPMI+10%FBS+100IU/ml IL2培养，最开始用cd3/28
beads active过，总是养几天之后越来越少，最后大部分都死光了！
哪位大虾有经验能给讲讲吗？多谢多谢！

t细胞3天以后要expand换plate，我们用的是plate bound的抗体，所以3天以后就没有
anti-CD3了

不好意思，我没有太看懂。
你是说你是用含有cd3的plate培养t细胞，每三天换一次新的plate是吗？
我是用cd3的beads培养的，应该也可以吧！
我只是不知道为什么会死

细胞密度是多少？naive T细胞培养必须维持高密度。太低就会死。

一般1-2million/ml

try add 2-mercaptoethanol in medium if you didnot add
The role of 2-mercaptoethanol in the stimulation of spleen cell cultures:
increased uptake of cystine into the TCA-soluble pool

太感谢了。一直有这个问题，养T细胞为什么medium里要加2-me。
