其他部分大同小异，贴一下关键的note部分
Note:
1. NgAgo/gDNA system is extremely sensitive to contamination of
intracellularbacteria (including mycoplasma) which are widespread
and leave no visible signs of presence. Carefully excluding the
presence of intracellular bacteria before performing experiments.
2. Because Agos need magnesium, avoid EDTA when detaching and seeding cells
into the plates for transfection. Alternatively, supplementing Mg 2+ to 5 mM
(may need optimization to your cell type).
3. Avoid using Lipofectamine® 3000 since the supplement P3000
interferes with ssDNA transfection. Lipofectamine® 2000 is a good
choice. Other
transfection methods such as electroporation are yet to be tested.
4. Ideally, 5' phosporylation of ssDNA guide by using T4 PNK (Biolab):

EDTA 是不透膜的，透膜的是EDTA-AM, 残存的那点EDTA早就被DMEM里的钙，镁给中和
掉了。
韩的protocal 转染之前还有一次换液，还剩多少EDTA在里面？

常用的 0.25% Trypsin with 1 mM EDTA
DMEM里含 2.4mM CaCl2, 0.8mM MgSO4, 干掉残存的那一丁点EDTA绰绰有余。
莫非alternatively那句话暗藏玄机？做不出来的赶紧去给培养基加点儿Mg试试。。。

这个 protocol 和 gluestic 六月28号发的三个tricks 中的两个能对得上。
看来 gluestic 和春雨有很近的联系。
下面是他当时的帖子：
Single stranded DNA is much more difficult compared to double-stranded
plasmid DNA in terms of transformation.
Try these single stranded DNA transformation tricks.
1) transformation done at a low pH of ~6. Low pH significantly reduces the
nuclease activity of cutting single stranded DNA.
2) increase Mg++ to 1 mM. Mg is a conformation stabilizer of DNA and RNA.
3) No EDTA in the transformation system because it deprives Mg++ from the
buffer.
Han is a seasoned scientist with expertise on nucleic acid research. He
knows all of these tricks and may not disclose all of them.
