老婆还不到24岁。。。准备12月初领证然后立即签证，因为化学的check较久，老板只
给了一个月假。没有合影。但是有我爸妈和她全家的多次合影。还有很多qq留言。求建

现在做蛋白分析， nanoFlow LC couple with LTQ orbitrap velos
我现在碰到的问题：
1）自己pack的column tailing很严重，是用买来的picofrit column pack的，压力
大概30bar左右。
2）MS/MS intensity特别低， parent ion intensity 有E+6， MS/MS intensity只有E
+2~E+3. 不知道是不是仪器出问题了。

判断仪器有没有问题很容易啊，拿calibration mix打一下看看，如果信号能达到E+7，
那机器可能就没有问题
会不会是样品的问题，样品的量本来就比较低。
buffer里加甲酸或许能减少tailing

1. 自己填充的柱子拖尾很正常，pack 柱子的技术含量挺高。你用nano柱，最好是买商
品填好的，重现性好。
2. ‘MS/MS intensity特别低， parent ion intensity 有E+6， MS/MS intensity只
有E: +2~E+3. ’这个在LTQ-Orbitrap上很正常，没问题。

给Thermo打电话了，人家的建议是1）check 是不是He气压力低了， 2）重新
calibration，可能由于误差， MS/MS的时候偏了。

有E: +2~E+3. ’这个在LTQ-Orbitrap上很正常，没问题。

关键是几个月前还有E4~E5的，最近突然低了几个数量级

1) The pressure is normal. Either the column or pre-column is contaminated.
Change a new pre-column, or check whether the packing material slurry or the
pressure packing device was contaminated.
2) What concentration do you have? If the intensity is too low for standard,
you might need to clean the ion optics ( transfer capillary, S-lens, etc).
Orbi Velos needs to be cleaned very often due to its flawed ion optics
design.

update一下，今天重新calibrate了系统， MS/MS intensity上去了
主要校正了transfer len系数
感觉orbitrap系统离子传送蛮tricky的
