There are two very important reports in recent Cell discussing how to design
potent siRNA.One is from Zamore lab, the other from Amgen company. Basically,
the rule is to put a weak base pair, for example A-T or even mismatch with
target, at the 5'-end of the antisense strand of siRNA. Such a design
faciliates RISC selecting the antisense strand as guide strand, instead of
sense strand. Most of previous failed siRNA sequences follow the opposite of
rule. Given this discovery, other considerations
