经过一周的讨论，公司现有两个职位
一个quality control lab scientist，65000. 主做分析。
一个inorganic synthesis chemist，70000. 主做合成。
责任差不多，先是学习自己lab所有的东西，然后manage技术员，然后开发新的东西。
我两方面都可以，主要是合成这块公司的东西，不论是材料，方法，还是装置，都和实
验室不一样。一开始去的话挑战大了一点！
此外，对于两个职位未来发展，大家如何看待？

看了realtor口头终止合同后,人家买了房子,她又跑来要钱的事,大家都很气愤,出了很
多主意.
心意是好的.但大部分不正确.可能会帮倒忙.我是一个realtor.因为我是非常喜欢这一
行,也是最成功的realtor之一,所以,我对于那些败坏realtor名声的害群之马,也深恶痛
绝.
但不能意气行事.要知道法律.
如果买主和realtor签了exclusive right to represent的合约,那么在合约结束之后的
60天内,realtor仍有权要commission.时间上各州不同.
买主的问题在于,当realtor口头终止合同时,没有文字记录.但是realtor有你签字的合
约.
因为绝大部分情况下,在美国买房,是卖主出commission.但是,若你的realtor不能从卖
主那里拿到commission, 她完全可以向你要.
目前的情形是,那个realtor给你下了个套,你钻进去了.如果上法庭,那么你输的机会相
当大.
唯一有可能成为证据的是,买主可能给后来的realtor讲过,原来realtor不愿再带他.如
果后来的realtor合作,法官有可能会采纳.real

，然后如何作用于473的？auto磷酸化的作用？
我们发现一个非PI3K依赖性的473变化，该如何解释呢？谢谢

是我的话，我会选inorganic synthesis chemist
公司里边做一些funtional 的东西如果出成果的话visibility 比较高一些。
quality control lab scientist 感觉晋升空间比较少，拿到的credit少，主要都是帮
别人干活了。

这个说的靠普。
其实很简单，op签了卖身契，然后没有半路终止的证据。

Generally, the kinases for T308 and S473 of Akt are PDK1 and mTOR complex 2
respectively. PI3K causes the production of PIP3 on the membrane, which then
binds to both Akt and PDK1 (which is assumed to be constitutively active).
So far there seems no direct evidence as to how mTORC2 is regulated. However
, inhibition of PI3K in most cases will lead to the suppression of
phosphorylation at S473.
Just curious, what is the evidence for PI3K-independent S473 phosphorylation
in your experiment? PI3K have different catalytic and regulatory subunits,
thus targeting PI3K genetically or via small molecules might not result in
loss of its kinase activity.

口头协议可以做证据，楼主可以先file small claim说agent违反合约。电话记录就可
以作证。

电话记录只是几点几分，通了多长时间的话，这个可以做口头协议的证据么？

我同意上位的观点，就是有可能在PI3K缺陷细胞或小分子抑制剂的情况下并没有完全
block其活性，我用了一个试验是在PI3K的p85缺陷细胞中检测到仍然有AKT473的调节，
但是否一定说明我这个调节是PI3K-independent的还为时过早。
为伊消的人憔悴。

If the buyer told several other people about this at that time, and these
people are willing to prove as witness, the buyer may have some thing to
argue about.

That seems more complex. The loss of PI3KR1 has been shown to cause
different or even opposite effects on downstream signaling. I would
recommend the use the p110 alpha or beta-null cells or specific inhibitor (
such as PIK75, TGX221) for your purpose. But it might not worth the effort
unless there is a potential to find out a new regulatory pathway or new
protein.

in which kind of condition the lost of p85 has opposite effect? especially
for p-AKT? i thought about p110, and anyone know the K/O cells avaiable?
