My boss wanna me do some NMR chemometrics research on -omics. Since my co-
supervisor is a mathematics professor and I am really into math and computer
science, I would like to make my project more mathematical. Do you think it
is a good idea for doing that, but the challenging part is I need to design
my project by myself well to combine both and balance both well.

1。你们如何解决样品库的来源?还是你们打算先用某些“训练样品”
来优化你们的算法?
2。我赞成你做更多数学方面的东西,这样你将来不想做生物了也比较容易
抽身而走,但是你可能得找一个得力合作者来给辅助你做生物那方面的东西。

我们也准备做这些。机器也有,都不太会玩儿。我们主要做海藻微生物代谢和lipid的

affiliated to our uni, some potential collaborations might be involved.
I quite like biology, but the wet experiment is just so tedious for me.

签证费最贵。美国收的贵,中国收的也贵,都是压榨血汗钱。

China just charges a few countries more, like US, Romania, because they
charge Chinese more. For all other countries, the charge is reasonable.
Every time US increases visa fee for China, China increases visa fee for
US too.
For the other question, you do not need to go to consulate directly to
apply for a Chinese visa -- you may let a friend or an agent help you.

there were two science papers, one was on human brain stem cells using
lipids as a stem cell/proliferation marker. I thought it is very cool and
has tremendous potentials but I have no expertise on the subject whatsoever
and cannot really comment on it...

I think that is where to apply statistical mode to discriminate molecules.
There is a metabolomics group in Imperial College Medicine School, they did
a lot chemometrics and NMR research on metabolomcs, quite famous in our
analytical chemistry filed.

从H1谱上来看,她们标记的不应该是脂类,因为化学位移只有1.28ppm,可能是某种烃

I don't know too much about the sensitivity of NMR. Actually MS sensity is
not good enough for many projects including mine. I was told the best MS was
around 20 attomolar.
How about NMR?
Another question: I guess for both NMR and MS in Metabolomics, it is not
widely used to characterize all the metabolites information in the sample. (
in contrast to microarray)
I read some paper and they always have a list of candidates to measure.
How long is the list of metabolites now?

我跟做有机的人聊过,nmr的sensitivity比MS要弱。不过因为是non-invasive的对特定
的实验用很好的用处。
至于代谢物list没有查过,估计在一百个左右的已知代谢物?
还有问题感觉就是定量,看过profiling的data感觉不少fold change并不理想,要在microarray数据里早就剃掉了

METLIN里很多不是代谢产物,
有MS/MS的是很小一部分.
Exact mass 也会给出多个可能.
与genonimcs不同, 代谢组学困难在如何鉴别. 我经常检测到几百个有变化的信号,
但最终能鉴别的只是几十个.
变化是很多,但你常常不知道是什么.

有MS/MS的是很小一部分.
Exact mass 也会给出多个可能.
与genonimcs不同, 代谢组学困难在如何鉴别. 我经常检测到几百个有变化的信号,
但最终能鉴别的只是几十个.
