不知道你是不是在说 bexarotene。恰恰相反,Gary E. Landreth在最初的文章[1]声 称bexarotene可以清除plaques---“Aβ plaque area was reduced more than 50% within just 72 hours.” 从而改善记忆,行为。但是被群起而攻之[2-5]。所有 的重复者都声称 no effects on amyloid deposition。其中的三个重复者声称 bexarotene对AD老鼠没有改善。只有一个重复者声称bexarotene对APP/PS1ΔE9老鼠的 记忆有所改善。之后Gary E. Landreth改口说: “We concluded back then that plaques don’t matter,” Landreth tells C&EN. “So it’s a mystery to me why these other investigators have focused on the irreproducibility of plaque removal. It seems like they’re beating a dead horse.” 究竟事情的真相如何,我就不知道了。不过我是不会相信Gary E. 不知道你在说啥?Bexarotene的文章提出了一个新的假设,用FDA已经prove的drug证明 了这个假设,而且有直接的clinical的相关性,因为这个药是已经FDA批准的了。 “We reasoned that an RXR agonist would enhance normal Aβ clearance mechanisms by activating PPARγ:RXR and LXR:RXR, inducing apoE expression, facilitating Aβ clearance, and promoting microglial phagocytosis.” 这篇nano的文章没有任何全新的理论,都是已经prove的,而且文章并没能给出 nanoparticle有什么advantage other than just an alternative.。 我个人觉得这篇nano除非有大牛支持,并且从一个新的角度重写,不然根本投不到CNS ,因为不对口,没有任何新意。冲击一下nat med或者sci trans med还有可能,但我不 是这个field,不敢妄加断言。
