my EB1a pp approved on May.
relink to NIW 485 and got cards on May.
But they sent me message and email update my status tonight to say "send you
approved notice on July 26th" although I got it month ago.
Anyway, seems they are working on EB1 cases

我想在一种肿瘤的细胞系中KD一个蛋白，用了两个不同的siRNA在多种细胞系中进行了
KD，结果qPCR检测显示所有的细胞系中KD效率都很好，但用了3种不同抗体的WESTERN检
测发现似乎没有KD效果。KD后有些细胞表型有改变，有些没有改变。请问可能的原因是
什么？

抗体的问题吧，要不是就是有些蛋白的稳定性高，你可以试试KD后72小时以上收sample

我试了3个公司的抗体，其中ABCAM的抗体背景很干净，但也没有KD效果。我也做了96小
时的KD，也没什么效果。。。

我被abcam的抗体坑过无数回，在right size会有条带，但是那条带不是你要的。我遇
见这种情况好几次了。感觉有些蛋白有好抗体，每家公司的抗体都基本work。有些蛋白
的抗体，没有一家的work。
