年初帮一个生物研究所里一个副头开发一个传感器（他们出钱），现在同一所里另一个
组头有兴趣把这个传感器（也许要改动设计）放到另一个应用上合作申请基金，不知道

我实验需要培养B6和一种mutant老鼠的Bone marrow macrophage,然后比较细胞的对LPS
,IFN beta， gamma等stimuli的反应区别，以及antigen presentation to CD4+ T
cell，nitro oxide production还有phagocytosis的区别。
我原来的protocol就是把bone marrow放在有M-CSF（L-Cell media）的media里养6-7天
，replate以后就用。后来隔壁实验室的老教授说，你这样养7天，得到的细胞其实更接
近于monocyte,还需要把细胞在no-M-CSF media里养2-3天，才能得到real macrophage
。可是我在文献中查不到这种说法的依据。
我老板原来是做T cell的，也不太清楚这个问题。
大家谁做过Macrophage啊？是不是如这个老师说的？还是说不同实验目的，培养方法也
不一样？

MCSF 6,7天出来的只要是贴壁的都基本上是MCs而不是monocyte了，他不信你让他去做

我们也是用Ｌ－ＳＵＰ刺激分化的。因为Ｌ－ＳＵＰ成份比较复杂，所以一周后去掉Ｌ
－ＳＵＰ再养几天比较好。对某些实验应该可以降低ｂａｃｋｇｒｏｕｎｄ。 这应该
对细胞分化没啥影响的。 我们用的是ＤＭＥＭ＋ｇｌｕｔａｍｉｎｅ， 没加其他的

是一周以后starve 的时候用DMEM+L-Glutamine么？还加horse serum 和Na pyruvate，
non-essential amino acid么？

如果需要ｓｔａｒｖｅ的话就不加ｓｅｒｕｍ， 如果不需要的话就加１０％的ｓｅｒ
ｕｍ（ＦＢＳ）

我是参照CURRENT PROTOCOL in immuology 的方法做的，和你的protocol差不多，只不
过我用的是 M-CSF,培养7天后flow cyotmetry显示有超过98%的细胞是CD11B+F4/80+,所
以他们是MACROPHAGE,不是monocytes。
