朋友硕士快毕业了，GPA比较高，所以收到这个入会邀请，需要交$95会费。请问参加
Golden Key International Honour Society对找工作有没有帮助？专业是土木工程的
结构工程。之前有八年相关工作经验，想在达拉斯附近找工作，但是工作不好找。有没
有什么建议？谢谢各位前辈！

LD是 primary petitioner，我以为我没有A number的。。。昨天找出了申请J2 EAD的
材料，发现移民局当时粘EAD卡的那个硬纸上面有个A#，不知道是不是就是本人的Alien
number？是不是应该把这个号码填到我的485申请里面。多谢！

Below are two SNPs I obtained for two samples from Exom DNAseq and Sanger.
From Sanger's results, it seems both SNPs are heterozygous, while GATK(v3.2)
call one sample as homozygous.
Does this mean we still need to do filtering based on sequencing depth,
which is hard filtering, then why do we need the machine learning-based soft
filtering?
If we need to do filtering based on sequencing depth, what threshold would
you recommend?
Sample1:GATK Sample2:GATK Sanger for both samples Sample1:IGV Sample2:IGV

这个A number 应该是1开头的，不应该填到485，485会有个2开头的A#

NGS has a much higher error rate. This is just a sequencing error, I think.
Trust the sanger result.

I-140申请approval notice只给了我LD主申请人A#，所以那我就是没有A#了？在485里
面就空着这格？

看一下vcf里每个位点的depth和quality score多少，可能太低默认扔掉了
sanger是怎么做的？光看peak overlap不能知道percentage，要准的话得做subcloning
然后测几十个克隆
