I was under the impression that perfusion is mainly to get rid of the blood
or more specifically IgGs out of the tissue so there will be much less
background in IHC.
Correct me if I am wrong
For embryos, endogenous IgGs are relatively low compared to adult tissues.
Maybe that's why it works well without perfusion.

我是觉得如果直接frozen tissue，剩下的tissue还可以做别的protein assay。要是
perfusion的话，就不能了。

perfusion with saline or PFA?
saline is to remove the blood from the tissue; and PFA is to fix the large
tissue that takes a long time to be fixed by immersion.

先saline把血洗了，然后perfuse with pfa。你会很明显地观察到一些特征表明各处基
本都fix了。。。这时候可以停了，取器官。。。

小鼠大脑切片，反正我们做的明显perfusion的比直接扔pfa的好看。
