depends on the size of cell aggregates. dual cells or triple cells are fine
for cell sorting. But big cell aggregates can't pass through and may get
machine jammed.

i found online that the lysate needs to be pre-filtered with 70um mesh
before sorting.
if the aggregates are smaller than 70um (say 60um), is it possible to sort?

I do use 45um filter to filter through the lysate before preparing the rest
procedures. I guess you can collect smaller aggregates less than certain
diameters.
The other thing is the purity of your cells. If the lysate is prepared from
heterogeneous population, where cells have different sizes, than you have to
be careful to isolate large cells and small aggregates.

你要tweak FACS才能收集aggregate，因为它default是要把所有不是single cell
event都排除掉的。此外cell aggregate形状不规则，你很难收集一个homogenous的
sample，会有很大的样本偏差。如果你规定只收集2 cell的，倒是还可能比较纯

俺不知道这个机器到底是怎么设计的，
能不能调整settings来sort性状比较规整的aggregates？
谢谢建议，下礼拜找管仪器的再问问。

100um nozzle works better for bigger cells or aggregates. You may simply
gate a region with the size you need. Size and shape are not the same, same
shape must have same size, or similar size is more likely to have better
uniform in shape.
