谁有xenograft的经验，帮我看看这张图啊# Biology - 生物学

I got interview in Chichago last friday. I made a presenation on my research
and 5 persons interviewed me afterwards. I spent 2 days writing thank you
letter. But only one person replied my email until now, saying I did good
job in the interview.
Does anyone know if this is good, or bad sign?

我在细胞系里knockdown了一个基因，然后注射到小鼠体内，形成肿瘤。肿瘤明显比对
照组要小。我们把肿瘤取出来，做切片，看ki67的表达。发现肿瘤切片里，有很多类似
colony的区域，ki67表达量很高，而他周围的区域相对较低。我感觉这些是knockdown
效率比较低的细胞，他们的生长比其他knockdown的细胞生长要快。但我看到对照组里
，也有这样的区域。感觉不太好解释。不知道谁做xenograft比较有经验，能给我讲讲
。特别是，我拍照的时候，应该选择哪个区域。谢谢了

貌似Ki67染色是nuclear的。不是片子干了吧？

In the image, the positively-stained area actually has less cells, thus
doesn't look like a colony. Also as aishang pointed out, the staining
pattern of KI67 is not typical.
For xenograft experiment, tumor size might be a more accurate readout (which
you already have). You can have plot, image of mice with tumor, image of
isolated tumors, western of tumor lysate to confirm the knocking-down. That
is already a bunch of data to show. Ki67 staining might be tricky, due to
highly heterogenous pattern you might see in a xenograft.

中间的染色有点问题。周围的是正常的情况。你是什么肿瘤细胞？

I think that is common too.
However, personally I think that is rude. In my previous job, my boss always
politely replied the candidates' messages, which made me quite impressed.
Actually when I was interviewed several years ago, he replied my thank you
letter too. Unfortunately, a lot of employers didn't do so, no wonder a lot
of Americans also complain of that, saying "employers, please stop playing
god!".

目前还不太清楚。但这是个膜蛋白，可能和integin有些联系。
