向大家求教如何验证chipseq中的antibody有效。做的是植物的histone modification
的ChIPseq实验，包括了几种mark，其中两种H3K9ac和H3K27ac的profile很像，所以
reviewer怀疑我们H3K27ac的antibody的有效性，要求我们给出相关的证据。因为本人
是做生物信息，不太清楚如何证明antibody的确实是有效的。希望大家能帮忙指点一二。

1. Pull down and then mass spectrum to check the target protein.
2. After ChIP experiment, load the sample to a protein gel, run western blot
with this antibody and other antibodies for same protein, include the other
antibody (H3K9 pull down, include h3k27, and vice versa) as well, to make
sure that you do pull down the right target but not the wrong target.

你的抗体是自己做的还是商业话公认的？
最严谨且变态的验证方法是敲掉k9me3的酶，没有了k9me3的pattern但27还在。同理

商用的，以前是用在人里头的，现在拿到植物里头来用。只是小文章，不想再做太多实
验了。

为啥要验证？商用的chip-seq ab, 把公司的ref翻出来，找找CNS把他拍翻

peptide blocking撒。。
你chipseq下来总要找几个target做chip-realtime验证吧。
side by side用对应的peptide block掉抗体，再realtime检测target。
比较快，大家也认可。
组蛋白修饰的抗体非特异的很多，前阵子还专门有人发了个nature structure xxx验证
了几十种抗体，哪些特异哪些不特异。
