标题: OPT转H4如何解释之前的unpaid intern

准备自己在美国境内转H4，之前的身份是F1的OPT，曾有几个月的时间做过一个unpaid
intern，当时和学校反复确认过这不违反任何法律规定。现在在填写I-539的时候，发
现需要填写有关之前所有employment的情况，并且要求写明薪水等信息，不知道我的这
个unpaid intern对转H4有没有影响？
希望高人指点！谢谢！

请问是否有应用Western Blot Imaging System的同学？我们目前有一台旧的Storm，因
为取消应用X-片，所以要买一台WB的成像仪，希望效果接近X-片,请各位大牛讲一讲你
们都用什么。

Almost all the modern western blot imaging system is better than films (
assuming you are using femto level substrate), I like the top of the line
alpha-innotech imager (do not get the red or blue one they try to sell; they
are horrible in my opinion). Bio-rad imager is very common but I think
their tech is the oldest. GE one is very nice. Do not get Kodak one if it
is still on the market (horrible).
Bio-rad recently join the ECL reagent market, their femto-ECL (Clarity) is a
lot cheaper than other companies (500 mL for less than $300) while Pierce
Dura is at 200 mL for ~$550.

we recently got FluorChem M from proteinsimple
I think it is capable of doing many different things, but it is a very slow
machine. I have no comparison with other machines.
the machine itself is fairly cheap than Bio-Rad and smaller.
it can be attached to a laptop, but the built-in computer is very slow with
Atom chip running Ubuntu.

Thanks for suggestions. GE Typhoon 9400 is very expensive, and the function
for detecting isotope is not a plus for us.

You don't need GE Typhoon at all, those are gel scanner. GE imagequant
LAS400 is the instrument you should look at. They are completely different
instrument. Unless you need to scan wet-gel with fluorescent/radio labeled
protein, you just need a camera based system not laser scanner.
Currently, I think Bio-rad, GE, and Alphainnotech (don't get their single
box instrument one - M, red, blue or whatever they call it-as mentioned
above; it is a horrible instrument) but their top of the line one running on
a PC is still my favor.

谢谢你的信息。太有用了。我正好最近也在找合适的。可不可以具体说说那一款
alphainnotech 的imager是你推荐的？我主要找detect chemiluminescence的imager,
别的功能不用太fancy,在保证质量的前提下越便宜越好。感觉是不是要在fluorchem的
系列里找， alphaimager应该不是你推荐的吧。另外Alphainnotech这个公司是不是现
在变成protein simple了？不好意思问题比较多。谢谢。

Search labx.com However, some of refurbished instruments are not
necessarily cheaper. I just brought a Tecan plate reader and the new one is
cheaper than the used quote and with more stuff in it.

Licor不是要用他们专门的二抗么，还有blocking buffer什么的。估计肯定超出我们的
budget了。

Thanks for the information. We tried Licor, but the sensitivity is worse
than film (we used result from film to compare with the image from any
machine for a demo). Currently, GE imagequant
LAS400 is the best, same result with film. The price is around $37000. The
Boss wouldn't try Alphainnotech, so please update if you try the machine.
Not sure if Bio-Rad is as same as GE LAS400.

one tube you could use for a year.
a little expensive, 50000?
I don't think it is less sensitive than film. and qualification is very easy
use its own software.

does it take for this one? 2 minutes? or 5 seconds?

曝光时间比较长，6 和12分钟
我看了下GE LAS 4000，似乎是原来的FUJIFILM LAS 4000，我们大组有这个系列的
MINI，记得不到1W刀。成像效果还不错。
我们实验室有过BIORAD 最新型号的DEMO，感觉比LAS 4000 MINI 强点但没有明显区别
，如果想省钱买BIORAD的DEMO，不到3W应该能下来了。

我们实验室最近也在用invitrogen的fluorescent 2nd abs，但是background很高，不
知道你们是多少稀释，或者说有没有什么trick？
