我要做一些蛋白DNA相互作用的实验，要求DNA终浓度大概在5uM。可是我做完PCR，纯
化之后算了一下只有大概0.25uM，差了20倍。请问一般如何可以提高PCR产物浓度？试
了提高引物浓度，好像不起效果。。。
请指点迷津！

100ul，100 cycles，taq, primer等都用终极的dose
不用hot lid
最后会浓缩到15ul左右，估计DNA浓度差不多了

pcr一般产物浓度在200nM就是不错的了。
主要是PCR的增加是exponential的，在20多cycle的时候多数都已经用光primer和dNTP
了。如果增加primer和dNTP的浓度，要知道，就需要exponentially increase。但是过
多的这些东西会抑制Taq的活性，所以一般跑30个cycle就到头了是有道理的。
如果实在需要那么大量的DNA，不如把PCR product给clone了，那就想要多少有多少了。

开checking不是hard，他们有另外一个system
40k TYP 可以在Expedia上用，可以换 $400 gift card (Staples, HD etc), 至少值个
$350吧

我觉得就算是美国人，也不会觉得不到150跟180+差别不大

Sure you can get the 20K TYP for $25k saving deposit. Same promo. The good
thing about citi's promo is you don't have to keep the money in the account
for a long time. As long as you did the intitial deposit, basically you
may withdraw it the 2nd day and still get the bonus.
