找工作一段时间了，大大小小的面试搞得身心疲惫。现在有一个工作基本搞定，对方明
确说面试过程结束，我是top candidate, 对方面世我的人也写了一封名为appointment
letter.本来我以为这个就是offer letter了，结果对方又发信要以前的supervisors
的references.
这本不是大事，但我头大的是，当初申请这个工作是通过学校系统投的，以前经验少的
时候我就把以前国内的研究生的经历写上。前俩月投工作的时候这个系统自动存的部分
我就没改。这下他们要我几个以前的supervisor的reference.
但是以前国内研究生时的老板肯定不会给我说好话（她要我读博士，我中途退了）。
我有没有办法跟这边说，提供别的supervisor 的联系方式啊？或者说那个supervisor
都是几年前的了，还在中国，我有更好的能说明我现在情况的supervisor
给我写？
多唐一句，这个工作本来面试过程就及其漫长和繁琐。现在找个工作可真难啊！小小
的一个学校里面的工作，面试就去了三次，又面试又考试的。而且面试一开始就已经
提供过三个referee乐，也都是我在

看到版上有人问了。我也查了下账户，没见到奖励点数。然后本来pending 在Nov 25的
两个bestbuy.com的单子，现在显示transaction date一个是26号，一个是30号，然后
post date一个是28号，一个是12月1号。
然后通过chase 的网站发信息去问，很快得到答复说我的purchase不eligible，因为不
在25号！然后说我们比较大方，尽管你不eligible我们还是决定奖励你点数XXX分。这
个奖励只给了第一个单子。第二个单子没提。
他们的回信说，当你通过chase ultimate reward mall的链接成功购买产品后，在4个
工作日内应该收到确认信。我想问大家都收到这个确认信了么？
注：为了防止链接不被承认。看好商品后，每次都是:
清历史，清cookie -> 关浏览器 ->打开浏览器（我用的IE9下单）->清历史，清cookie
->登陆chase网站 -> 通过他们的链接到商家 -> 选好商品用chase sapphire结帐。
中间都是一次操作。基本上除了重启电脑外，能清的都清了，能关的都关了。然后还告
诉我不eligible，这太让人费解了。

最近在做一个研究转录因子的靶基因课题，用siRNA后48小时，蛋白只剩了不到10%，
然后用Affimatrix array比较non-target control siRNA knock-down 对照后，看可
能的靶基因。结果只有几十个基因变化大于1.5-fold。
是人的primary cell, 大家有啥好建议，谢谢了。

以下是他们的回信，他们竟然说用Win7有可能造成无法顺利得到分数，是不是有些夸张
呀？
不过我倒是确定我购买的时候绝对是一个tab内完成的。没有开多个tab。怎么还不
eligible？
是bestbuy.com改了大家的pending还是chase改了transaction？让人费解@@

没做过。瞎说两句。
你为啥看蛋白，而不是mRNA的最低点？
你这个TF是已知很多靶基因么？会不会本来就只调控几个基因的
有没有ChIP的data?

唉，那你准备怎么解决呢？实在不想把以前那个supervisor供出去，现在手头已经积攒
了几个可以提供reference的人，哪个都比这个好。。
只有恨自己懒，当时投的时候也没有改改系统里的信息。

我得有确认信！不过估计大家post day肯定都不是11／25,那天bb 根本忙不过来。我
得statement 今天出来，看看有没有。没有就打电话，应该能拿到地。。。

因为最后工作的是蛋白，我也看了mRNA，有4倍的down-regulation
不知道有啥靶基因，我已经做了ChIP，所以想用siRNA结果来证明。
同时在做 KO mice, 但是拿老鼠的结果跟人的比总归不太好。

没有确认信，应该是BB改了post day，我的现在也是12/1号了，点数有可能更没希望了

1.5 fold好像太小了。是用fibroblast吗？一般我们Affy都能得到几百上千个变化超过
2倍的。
还有一种方法是做KO human cells，比如用Talen.

不好意思，弱问一下怎么查账户的奖励点数啊？进去chase自己的账户后点哪个按钮啊
？我25号也在bestbuy买了1000美元的东西，希望能有他们说的那10% back奖励点数。

this is very common. if your TF is stress related, you probably have better
chance to see more genes show higher changes. You can check all the
predicted targets of this TF and see whether there is a systematic
differences between these putative target genes, since most of those target
genes may not show significantly changes if you look at them one by one.

可他们真想查，网上艘艘总能搜出来，而且他们实验室有中国人，如果中文一搜就更。。
已经查过我三个referee乐，谁知道还要查。心里没有底。sigh..

之前下过两单sephora
没有确认信
问了客服也说没有eligible purchase
然后statement出来时候两单都给了。。。

If the TF is stress related, can you treat the cell with stress, and then
analyze the transcriptome changes?

我大大小小面世过来，第一次遇到那么严谨繁琐的，我最当前的三个supervisor 都已
经写过reference 啦。。我也去考过好几次了。又来。。
