第一份工作是一家外资企业，工资4000，加上餐补，加班，估计就3300多点，因为受
不了英语，虽然英语过了CET-6，但由于接触的有些是专业术语，同事大部分在美国，
要邮件联系解决问题，干了几天就不想去了。第二份工作是一家台湾企业，做自行车车
架的，材料什么很繁琐，负责应付，干了三天，发现自己不能适应就散人了。第三份工
作是家上市公司，因为同事的一句话，“你简直是人才”，索性不去了。这换工作的频
率估计也只有我才做的出来，其实心里都明白，职场毕竟不是校园，很多地方，很多压
力都要自己去克服，自己也不知道是因为没有做好准备工作，还是因为对自己的不信任
，可能更对的是对职业的迷茫吧。不知道自己到底适合干什么了！而且找的工作是工资
一份比一份低，现在想来还是第一份工作最好，有我喜欢的英语，还是会计专业，同事
也好相处，可是就是因为自己的冲动莽撞，不愿意尝试导致现在的结果。接下来我是更
迷茫了、、、、、、

Note: NHEs is membrane proteins that transmembrane 12 times. NHE5 is
localized to the plasma membrane;NHE6-9 reside on organellar membranes(
endosome,Golgi);
1. pcDNA transfects NHE5-9 through Fugene 6(ratio is 3 uL of Fugene/2 ug of
DNA) in 293 cells. After three days,collect cells.
2. Cold PBS washes cells. Add 150 uL of lysis buffer(final concentration:
50mM Tris pH7.4, 150mM NaCl, 1% triton, 0.1% sds, cocktail inhibitor)
into each well of 6-well plate. Scrape cells.
3. The lysate gets sticky right now. Use syringe sucks up and down by 10
times,or sonication; Keep in ice for 20 minutes;
4. Then add 4x sample buffer (the final concentration of sample buffer is:
50mM Tris, 1% sds, 5% glycerol,10% beta ME). Set at room temperature for 30
minutes. No boiling, because nothing can be detected in Western blotting
once boiling. Heat in 37 degree,or 70 degree for 10 minutes, the ECL signal
of blotting decreases over heating temperature;
5. Run SDS-PAGE gel( bio-rad precast gel,4-15% tris-HCL,15 wells,cat #161-
1122) at 100 volts for 1 hour; wet transfer at 100 volts for 1 hour;
6. 5% BSA in TBST for 30 minutes. Rb Flag tag antibody(cell signaling,
cat #2368, 1:10000 in 5% BSA) for 3 hours; Wash 3 times,10 minutes each. 2nd
antibody is 1:10000 for 1 hour. ECL ,develop.
7. Look at the film attached. The over expression is successful,compared
with empty ctrl in the left lane(pcDNA transfection without NHE). But always
can't find any sharp band in 70-100 kD target region. The positive lane is
black starting from top. Seems proteins can't separate well. Aggregation is
too much? So what happened?
a. NHE overexpression has some problems?Change Fugene:DNA ratio?
b. Lysis buffer recipe is wrong, add more SDS to digest member protein?
incubate longer?
c. sample buffer need more beta-ME? incubate longer?
d. change dilution ratio of 1st and 2nd antibody?

现在基本上是个显卡都至少支持两个.

支持两个显示器和支持三个是完全不同的概念，显卡上的花费也差数倍，不是有三个头
就能同时输出到三个显示器，这个要考虑清楚

带display port的HD5xxx以上支持三屏，详情google一下eyefinity
其实不如买俩亮机卡合算

我是有个独立的显卡，然后一个是VGA, 一个是DVI，分别接在2个显示器上，但其实显
卡上还有个
HDMI的接口，那个HDMI的接口还能接地三个显示器？
还是得再买个显卡，然后把第三个显示器接在那？

得另买个卡，我正用着三台显示器

买2个显卡能支持三个显示器吗？我是有个独立的显卡，然后一个是VGA, 一个是DVI，
分别接在2个显示器上，但其实显卡上还有个HDMI的接口，那个HDMI的接口还能接地三
