最近在考虑换门窗。 请问有没有人用过恒益门窗的？ 从他们的网站和介绍看起来还不

3个月前，种的光脚梅/李树，现在还都光秃秃的，梅的主杆和枝条还都是绿的，掰它们
还有弹性，，李的主杆有点绿，但枝条都干了，，等了这么久，一点要萌芽的迹象都没
有，，树神们告诉我，还需要再等吗？还要等多久？？
还有3棵光脚玫瑰，同期的都发芽长花苞了，一棵干脆黑了，就它们杆还绿着，但纹丝
不动如初，，这还要等多久？玫瑰党的筒子们来说说。

最近在做Chromatin IP, 以前都做转录因子结合DNA，一般先做个预测，然后蛋白拉
下来做pcr。老板说想看下转录因子结合后，组蛋白在该基因启动子区域结合的变化。
我想请教一下组蛋白结合在DNA启动子上的什么区域，好方便设计引物？是ATG
start codon附近还是TATA box之类的呢？
另外，如果我选一个促进的，一个抑制的组蛋白，可以选H3K9Ac和H3K9Me2/
H3K27Me2的antibody吗？
以前没有做过组蛋白的问题，请熟手不吝赐教，感激不尽。

你是打算做seq还是pcr? seq的话所有基因都看到，pcr的话你怎么做
定量？realtime? 另外，组蛋白是everywhere，除了start site低一点，
你怎么设计引物？还是你打算看特定位置的组蛋白，比如-1?

them as they do not do Aluminum windows.
I pick American Vision + Milgard window....it is really hard to get a good
price from American Vision, but they have skilled workers and good attitude
in all the process...good customer services too. I recommend AV, but you
have to be patient to negociate a good price with them...the first quote
they gave me was $7800 for 11 windows. My final deal is $3800. The first and
final quo

每个nucleosome里面组蛋白是个octamer，h2a, h2b, h3, h4 X2，外面饶130左右个bp

知道你能找到好的抗体不？印象中Me2，Me同一个抗体都识别。还有你怎么确定这个H3
的PTM只有你要的K9 Me2 or Ac？

H3K9Me2和H3K9Ac不是组蛋白，只是组蛋白H3尾巴上的某个氨基酸的修饰。针对这些修
饰有特异的抗体（当然有的不像自己声称的那么特异）
选K27me3/2 K9me3/2/1 K4me3/2 K9ac看看，再用针对H3的抗体做control，normalize
在TSS前边选几段 做Q PCR
还可以看H2Bub，pol II的磷酸化吧，根据你的实验和对细胞的处理
