今天受到email据了AP EAD. 上网查状态140 485都批了...thank lord

laken here in this board helped with letters and petition letters, letters
improved a lot from 3 years ago's NIW letters

最近用WB检测一个蛋白，分子量140KDa，经常出现如上图所示的情况，就是主带很糊，
而且主带下方总会有一条细带，孔与孔之间互相连接，做其他小分子的蛋白从来没遇到
这种情况，请问到底是哪些原因？求大神建议。谢谢

好几个原因丰度高上样体积太大堆积胶略微有点缩水导致和玻璃板之间略微有点脱
离堆积的时候有些样品会从玻璃和胶之间直接渗到堆积胶和分离胶的界限上出现这种

有人甚至怀疑过制胶板污染。当然，可能性极小，因为还要求大小一致，但也是可能性
之一。LS所说的从胶和板之间走样，我也是很难理解的，因为我想这种走法几乎没有电
阻，不会与胶内走的样品跑在一条线上的。
最后，觉得曝光时小幅度内移动，特别是你放胶片下去的过程中，在没与膜接触之前，
光片已有曝光，就会造成这种鬼影。你只要降低上样量，就可解决。

刚开始跑的时候你看看loading有没有一点点沿着孔的角向两边漏？这样跑的会比在孔
里快一点最后爆出来就是这个样子
把浓缩胶的tris buffer ph严格调了一下就好了。
你可以把loading弄的蓝一点跑跑看，这样比较显眼
不是从胶和板之间走样，还是在胶里面，我剥开看过的。

tris buffer ph 很重要的。

制胶后都水封啊。如果制的胶新鲜，应该不会有缩水问题。否则就是架子不给力啊！

同学，倒完分离胶后，貌似不该用水封，该用1-butanol (用水饱和的，醇在上层）。
用水封很容易破坏界面甚至是顶层胶的结构，影响跑胶结果

倒完分离胶后,我们常规用isopropanol封顶的。
