我们在小鼠里敲除一个基因后，做TUNNEL assay,发现retina的inner nuclear layer呈
阳性（上图箭头处），而control则是阴性（下图箭头处）。 但有个同事说是假阳性，
因为真正的阳性细胞应该看上去偏小，呈小点状，而且组织结构应该会mess up,但我这
个retina，看这结构还很完整，不像apoptosis.大家说呢？谢谢。

你没看见上面【提示】就有吗？倒数第二个【提示】就是答案啊！

What is your control, Wild type mice control or no antibody control?
It does not looks positive from my view. It is unlikely that all of these
cells in this layer are uniformly positive. In addition, the cell size of
positive cells in your picture look pretty round and uniform, while the size
of true positive apoptotic cells are quite heterogeneous and look different
from healthy cells.

谢谢！是wild type mice control.你说的和我同事说的很接近。但是我搞不懂，为什
么control这么干净。我两个是平行做的。

这个到是没做，需要一个inner nuclear layer undergoing apoptosis的control...
不过我在网上找到这个（见贴图），确实如spt009说的，和我的有些不一样。
TUNNEL是用来stain DNA nick,也许在我的sample里，可能不是apoptosis,但确实DNA
fragmentation增多了。

If it is wild type littermate control, it is worthy to repeat more mice to
see whether the staining is reproducible. If yes, maybe there is something
going on.

直接用DNase做positive control.
这样可以告诉你至少两个信息。
1 你的TUNNEL工作了没？
2 阳性对照下的morphology。
上面说的对，你需要多重复重复。
不过在有tunnel信号的时候，组织不一定会mess up.
