《自然—细胞生物学》:清华大学俞立团队揭示新的血管新生调控机制
北京时间2022年11月29日,清华大学俞立教授团队在《自然—细胞生物学》上发表题为“Monocytes deposit migrasomes to promote embryonic angiogenesis”的研究论文,从全新的角度阐述血管新生机制。
一个偶然的机会,研究者观察到鸡胚绒毛尿囊膜(CAM)上存在一群高度迁移的细胞,经鉴定,该类细胞为单核细胞,在鸡胚尿囊膜上产生大量迁移体,看上去就像孔雀开屏(图一:单核细胞迁移时产生迁移体)。去除单核细胞导致毛细血管形成受损,原因是单核细胞产生的迁移体能够诱导血管新生。敲低或敲除TSPAN4阻断单核细胞迁移体的形成,继而抑制毛细血管新生。同时,单核细胞产生的迁移体可以在体外和体内募集单核细胞。
研究者进一步发现迁移体促进血管新生的机制是迁移体高度富集CXCL12和VEGFA。迁移体分别通过释放CXCL12和VEGFA来募集单核细胞和促进血管新生。
